CAS number: 818-15-5,
1,2-Diamino-4,5-methylenedioxybenzene (DMB) is a versatile fluorescent labeling agent. DMB is currently manufactured by Hixia Custom Synthesis in 100g quantities and the material produced is primarily used in biological applications such as fluorescent labeling of sialic acids. Sialic acids are a family of 9-carbon carboxylated 2-keto sugars found on many glycans associated with glycoproteins. Over 25 sialic acids have been identified in nature and have been shown to play a major role in a variety of biological situations.
Application:
1,2-diamino-4,5-methylenedioxybenzene (DMB) is used in the fluorescent labeling of sialic acids. Sialic acids released from glycoconjugates have been identified by a variety of analytical techniques including: TLC, GC, GC-MS, FAB-MS and HPLC. Of these techniques, derivatization of sialic acids with 1,2-diamino-4,5-methylenedioxybenzene (DMB) prior to separation by reverse-phase HPLC offers an excellent analytical method with many advantages (1,2,3):
High sensitivity and specificity
Good resolution of isomers
Minimal artifacts due to O-acetyl ester migration from the 7- to 9-position
Detection: The conjugated 1,2-diamino-4,5-methylenedioxybenzene (DMB) labeled sialic acids have the following fluorescence characteristics :
l ex = 373 nm, l em = 448 nm
